Vascular Endothelial Growth Factor-A (VEGF-A) was originally isolated from tumor cells and is produced by a wide variety of cell types. In addition to stimulating vascular growth and vascular permeability, VEGF-A may play a role in stimulating vasodilatation via nitric oxide-dependent pathways. VEGF-A has several variants, VEGF-165 being the most abundant. Rat and bovine VEGF are one amino acid shorter than the human factor, and the bovine and human sequences show a homology of 95%. Recombinant rat VEGF-165 is a disulfide linked homodimer, containing two 165 amino acids chains, with a total molecular weight of 38.8 kDa.

SDS-PAGE of Rat Vascular Endothelial Growth Factor-165 Recombinant Protein. Lane 1: 1 ug Rat VEGF-165 in non-reducing conditions (-). Lane 2: Molecular weight marker. Lane 3: 1 ug Rat VEGF-165 in reducing conditions (+). Rat VEGF-165 is predicted to be a homodimer with a total MW of 38.8 kDa.
Bioactivity of Rat Vascular Endothelial Growth Factor-165 Recombinant Protein. Serial dilutions of Rat VEGF165, starting at 250 ng/mL, were added to HUVECs. After 87 hours, cell proliferation was measured and the linear portion of the curve was us used to calculate the ED50. The ED50 of Rat VEGF165 is 5.9-8.9 ng/mL. This value is comparable to the typical expected range of 4-8 ng/mL.
