Transforming growth factor beta isoforms (TGF-beta1; 2; 3) play vital roles in regulating cellular growth and differentiation; and they signal throμgh a highly restricted subset of receptors known as TGF-beta type I receptor (TbetaR-I) and TGF-beta type II receptor(TbetaR-II). TGF-beta's specificity for TbetaR-I has been proposed to arise from its pre-helix extension; a five-residue loop that binds in the cleft between TGF-beta and TbetaR-II. TGF-beta binds with high affinity to the type II receptor; a transmembrane protein with a cytoplasmic serine/threonine kinase domain; which requires both its kinase activity and association with another TGF beta-binding protein; the type I receptor; to signal growth inhibition and early gene responses. Receptors I and II associate as interdependent components of a heteromeric complex: receptor I requires receptor II to bind TGF beta; and receptor II requires receptor I to signal. Mutations in TbetaR-I and TbetaR-II gene have been associated with Loeys-Dietz aortic aneurysm syndrome (LDAS).
