PROGEN’s Comprehensive Workflow
The hyperphage production stage involves engineering E. coli to produce hyperphages lacking the pIII protein, using a helper phage vector (M13KO7ΔpIII). This system allows precise control over which proteins are displayed on the phage surface.
In the hyperphage display stage, antibody fragments (scFv) are cloned into a phagemid vector containing a pIII gene. This construct is introduced into E. coli, which, when infected by the hyperphage, produces phages displaying the scFv–pIII fusion protein on their surface.
During panning and elution, these phages are exposed to an antigen to identify strong binders. After washing, the bound phages are eluted using trypsin and re-amplified in E. coli. After 2–3 rounds, high-affinity binders are isolated and can be cloned into various formats for production of recombinant antibodies.
