mBDNF vs. proBDNF
The Yin and Yang of Brain Health: mBDNF vs. proBDNF
Brain-Derived Neurotrophic Factor (BDNF) is often described as a key regulator of brain health. It exists in two primary forms mature BDNF (mBDNF) and its precursor, proBDNF, which can exert distinct, and in some contexts opposing, biological effects.
Understanding the relationship between these two isoforms is increasingly important in research into depression, Alzheimer’s disease, synaptic plasticity, and neurodegeneration. However, because they are structurally related, measuring them individually can present a significant technical challenge.
Distinguishing mBDNF from proBDNF in Research
It was once thought that proBDNF served only as an inactive precursor that required cleavage to generate biologically active mature BDNF. It is now understood that proBDNF can also function as a signalling molecule, often influencing pathways distinct from those associated with mature BDNF.
Feature | Mature BDNF (mBDNF) | proBDNF |
Primary Receptor | TrkB (Tropomyosin receptor kinase B) | p75NTR (p75 neurotrophin receptor) |
Main Function | Neuronal survival, differentiation, and growth. | Promotes apoptosis (cell death) and axonal retraction. |
Synaptic Impact | Enhances LTP (Long-Term Potentiation) | Enhances LTD (Long-Term Depression) |
Research Focus | Plasticity, recovery, antidepressant response | Neurodegeneration, stress, synaptic pruning |
Why the Ratio Matters
In healthy systems, there is often a balance between mature BDNF and proBDNF. In some pathological conditions, this balance may shift.
Research suggests that altered proBDNF:mBDNF ratios may be associated with cognitive decline, mood disorders, injury responses, and impaired synaptic function. Altered processing of proBDNF into mature BDNF has been proposed as one mechanism that may contribute to these shifts.
For many researchers, measuring both isoforms and not simply total BDNF, can provide a more informative view of BDNF biology.
The Challenge: Measuring What Matters
Some conventional BDNF assays may not adequately distinguish mature BDNF from proBDNF, potentially confounding interpretation.
Because the mature BDNF sequence is contained within proBDNF, assays lacking sufficient specificity may detect overlapping signals, making it difficult to determine whether changes reflect mature BDNF, proBDNF, or both.
Precision Tools for BDNF Research
To solve this, Biosensis has developed a suite of Rapid ELISA Kits designed to give researchers absolute clarity on which isoform they are looking at.
1. Human mBDNF Rapid ELISA (BEK-2211-2P)
The Human mBDNF Rapid ELISA is optimised for preferential quantification of mature BDNF, with low cross-reactivity to full-length proBDNF. This assay is well suited for studies focused on synaptic plasticity, neuroprotection, exercise-induced BDNF biology, and broader investigations into neurotrophic pathway modulation.
For researchers specifically interested in measuring mature BDNF-associated signalling, this assay provides a targeted solution for isoform-specific analysis.
2. Human proBDNF Rapid ELISA (BEK-2237-2P)
The Human proBDNF Rapid ELISA is designed to detect full-length proBDNF through recognition of the pro-domain, with minimal mature BDNF cross-reactivity. This assay supports research into neurodegeneration, stress signalling, synaptic pruning, and mechanisms related to BDNF processing.
For studies focused on precursor biology and pathways associated with neuronal vulnerability, this kit provides a dedicated tool for proBDNF measurement.
3. The mBDNF / proBDNF Combo Set (BEK-2241-SET)
Combines complementary mature BDNF and proBDNF ELISAs, enabling measurement of both isoforms across the same sample cohort.
By supporting analysis of BDNF isoform balance, the combo set can provide deeper biological insight than measuring either form alone.
Don’t Measure One Isoform in Isolation
Studying mature BDNF or proBDNF alone may provide only part of the picture. For researchers investigating BDNF processing, neurotrophic signalling, or shifts in isoform balance, measuring both may offer a more complete understanding of the biology at play.
