- VISA is an important protein required for innate immune defence against viruses. VISA acts downstream of DDX58 and IFIH1/MDA5, which detect intracellular dsRNA produced during viral replication, to coordinate pathways leading to the activation of NF-kappa-B, IRF3 and IRF7, and to the subsequent induction of antiviral cytokines such as IFN-beta and RANTES (CCL5). VISA is believed to activate the same pathways following detection of extracellular dsRNA by TLR3 and it has been hypothosized that VISA may protect cells from apoptosis. VISA is a ubiquitously expressed protein present in a range of cell types including T-cells, monocytes and epithelial cells, highest levels in tissues of the heart, skeletal muscle, liver, placenta and in peripheral blood leukocytes. Multiple isoforms of VISA have been identified. Applications: Suitable for use in ELISA, Immunofluorescence, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Immunohistochemistry (Paraffin): Requires antigen retrieval using heat treatment and sodium citrate buffer, pH 6.0, prior to staining of paraffin sections. Optimal dilutions to be determined by the researcher. Immunohistochemistry Positive Control Tissue: Mouse brain Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
