- UBASH3B is a cytosolic 72-74kD member of the TulA (T cell ubiquitin ligand) family of proteins. It is widely expressed and appears to perform multiple functions. As a histidine phosphatase, it dephosphorylates Syk and Syk-related molecules, thus dampening signaling pathways. Via ubiquitin and SH3 binding domains, it binds to monoubiquitinated RTKs and Cbl, thereby increasing RTK activity by blocking its polyubiquitination and subsequent lysosomal degradation. There is also the potential for phosphodiesterase activity directed towards tRNA activity. Human UBASH3B is 649aa in length. It contains one UBA domain (aa39-75), a proline-rich ligand binding SH3 region (aa257-316), and a PGM/histidine phosphatase domain (aa386-598). UBASH3B is suggested to dimerize. Over aa47-253, human and mouse UBASH3B are identical in aa sequence. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 2ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
