- The thyroid microsomal antigen has been shown to be the enzyme thyroid peroxidase (TPO). TPO is a membrane-bound, glycosylated, hemoprotein enzyme with a molecular weight of approximately 110kD. It plays a significant role in the biosynthesis of thyroid hormones by catalyzing both the iodination of tyrosyl residues in thyroglobulin and the coupling of the iodotyrosyl residues to form thyroxine (T4) and triiodothyronine (T3). Autoantibodies produced against TPO are capable of inhibiting the enzyme activity. They are also complement fixing antibodies that can induce cytotoxic changes in cells and are, therefore, a potential cause of thyroid dysfunction. The measurement of low levels of TPO antibodies in serum can be useful in the assessment of a number of thyroid disorders. More than 90% of patients with autoimmune thyroiditis (Hashimoto's thyroiditis) have thyroglobulin or TPO antibodies. While not diagnostic, the detection of TPO antibodies can aid in predicting the progression of chronic thyroiditis and in further substantiating thyroid disease in patients with non-thyroidal illness. Antibodies to TPO have also been found in most patients with idiopathic hypothyroidism (85%) and Graves' disease (50%), and less frequently in patients with other thyroid disorders. Low titers may also be found in 5-10% of normal individuals. Applications: Suitable for use in ELISA, Immunoelectrophoresis and Affinity Purification. Other applications have not been tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Hybridoma: SP2/0 myeloma cells with spleen cells from Balb/c mice. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
