CTEN, also known as Tensin 4, is a 90kD cytoplasmic protein that is associated with focal adhesions in epithelial cells and is upregulated in several carcinomas. It promotes tumorigenesis by disrupting the attachment of focal adhesions to the cytoskeleton, promoting epithelial-mesenchymal transition, and binding nuclear beta-Catenin. CTEN is cleaved by Caspase-3 at aa570-571 during apoptosis. It contains a serine-rich region aa168-324, one SH2 domain aa449-556, and a phosphatase domain aa548-714. Within aa1-107, human CTEN shares 62% and 59% aa sequence identity with mouse and rat CTEN. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml Immunohistochemistry: 8-25ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
