Human Secretory Leukocyte Protease Inhibitor SLPI is a protein found in various secretory fluids, including parotid secretions, bronchial, nasal and cervical mucus, and seminal fluid. The protein is a potent inhibitor of a variety of proteolytic enzymes, including the neutrophil proteases, elastase and cathepsin G and the pancreatic proteases chymotrypsin and trypsin. SLPI is an important component of the antiprotease defense of tissues bathed by secretory fluids and could be useful as a therapeutic agent for treatment of the proteolytic damage of tissues seen in degenerative and inflammatory diseases. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 0.1-0.2ug/ml with the appropriate secondary reagents to detect human SLPI. The detection limit for rhSLPI is ~1ng/lane and 5ng/lane under non-reducing and reducing conditions, respectively. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
