- Rheoviridae/Rotavirus/Rotavirus A/Simian rotavirus SA11. Virions consist of a capsid, a core, and a nucleoprotein complex. Virus capsid is not enveloped. Capsid/nucleocapsid is isometric with icosahedral symmetry and has a diameter of 80 nm. The genome is segmented and consists of eleven segments of linear double-stranded RNA. Rotavirus is the most common cause of severe diarrhea among children. The incubation period for rotavirus disease is approximately 2 days. The disease is characterized by vomiting and watery diarrhea for 3 - 8 days, and fever and abdominal pain occur frequently. Source: Rotavirus, SA-11 strain, cultured in MA104 cells. Activity: Activity of the product was evaluated using a lateral flow rapid immunoassay and the signal intensity generated by a test antigen was compared to the single intensity of the reference material at the same dilution. Inactivation: Gamma irradiation. This procedure is effective primarily by damaging viral genetic material. Rotavirus inactivation is assessed using a double-passage assay in tissue culture. Rotavirus antigen is inoculated onto an MA104 cell monolayer and incubated for three days. The infected culture is passaged and scored after a further 3 days of incubation for infectivity. The culture is monitored for cytopathic effect. An absence of cytopathic affect, within the limit of the assay, indicates the inactivation of the antigen. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70°C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
