- Raf-1 is a ubiquitously expressed 74kD member of the serine/threonine protein kinase family. Raf-1 is a MAP kinase kinase kinase and participates in the transduction of mitogenic signals from receptors on the cell membrane to the nucleus. Raf-1 is activated through phosphorylation by Ras and in turn phosphorylates MEK which activates ERK. ERK when activated regulates the expression of genes involved in cell cycle, differentiation, migration and apoptosis. As such Raf-1 is an important clinical target in cancer. Raf-1 activity can be lowered by phosphorylation at serine residues 301 and 642. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
