- Poly [ADP-ribose] polymerase-1 (PARP) is a nuclear protein that catalyses the poly ADP-ribosylation of a limited number of acceptor proteins involved in chromatin architecture and in DNA metabolism. It is activated by DNA strand breaks and plays a role in DNA-base excision repair, DNA-damage signalling, transcription regulation and proteosomal function. PARP is cleaved by caspases during apoptosis to yield two apoptotic fragments, the amino-terminal DNA binding domain (25 kD) and the carboxy-terminal catalytic domain (85 kD). PARP cleavage facilitates the cellular disassembly and serves as a marker of cell undergoing apoptosis. Applications: Suitable for use in Western Blot and Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Immunocytochemistry: 1:100 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
