- PAK3 is a cytoplasmic 68kD member of PAK group I, STE20 subfamily, STE Ser/Thr protein kinase family of molecules. It is expressed in neurons of cortical layers II, III and V, the arcuate nucleus, and neurons in the dorsal raphe plus locus coeruleus. PAK3mediates axon and dentrite arborization, providing volume to the mass of the CNS. It binds to active GTPases which induce PAK autophosphorylation and activation. Human PAK3(b) is 559aa in length. It contains one GTPase binding domain aa68-128 plus a protein kinase catalytic region aa283-534. Acetylation occurs on 535aa, and there are at least three potential phosphorylation sites. Alternate splicing generates three isoform variants. There is a 65kD short form that shows a deletion of aa93-107 (PAK3a), a 69kD long form that shows a 21aa substitution for aa93-107 (PAK3c), and a 72kD very long isoform that contains the aforementioned 21aa "substitution" inserted onto PAK3b after Thr92 (PAK3cb). PAK3b, c and cb are all constitutively active, and show only modest binding affinity towards GTPases. Over aa138-255, human PAK3(b) shares 94% aa identity with mouse PAK3(b). Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.25ug/ml Immunohistochemistry: 5-15ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
