Several peptides associated with feeding behavior have been reported recently. Orexins (Orexin-A and Orexin-B) are a family of hypothalamic neuropeptides selectively expressed in the hypothalamus (1). Orexin-A and Orexin-B are derived from the same precursor (Prepro-orexin) by proteolytic cleavage. Prepro-orexin is 130 amino acid long peptide with a putative 33 AA secretory sequence, a hydrophobic core followed by residues with small polar side chains. The expression was detected in brain and to a small extent in testis (1). These neuropeptides bind and activate two closely related Orexin receptors—G-protein coupled receptors (GPCRs) OX1R and OX2R. Applications: Suitable for use in ELISA. Western Blot, though not tested, may potentially be used as an application. Other applications not tested. Recommended Dilution: ELISA: 1:10,000-1:100,000 Using 50-100ng of control peptide/well. Western Blot: 1:1000-1:5000 Optimal dilution determined by the researcher. Control Peptide: O7501-10 Storage and Stability: Lyophilized powder may be stored at 4°C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Specificity:
Recognizes full-length (1-28aa) Orexin B. The epitope is not found in Orexin A. The appropriate control immunogenic peptides may be used to confirm specificity of antibodies. Since Orexins A and Orexin B are derived from an unprocessed pre-pro orexins (130aa), the anti-Orexin B antibodies may also react with prepro orexins. Species Crossreactivity: Mouse, rat and human.
