- Human 8-Oxoguanine DNA Glycosylase (hOGG1) is involved in the excision of 7, 8-dihydro-8-oxoguanine produced as a result of DNA damage. This gene has been localized on human chromosome 3p25. Several forms of hOGG protein have been cloned differing in lengths (351-424 aa) (refs 1). Full length human OGG1 fusion protein with N-terminal His-tag was expressed in E. coli and purified using Nickel-affinity support. Purified hOGG1 was injected into rabbits to produce antibodies. Applications: Suitable for use in ELISA. Western Blot, though not tested, may potentially be used as an application. Other applications not tested. Recommended Dilution: Western Blot: 1:1000-1:5000. Deytects a band at ~38kD from human cells and tissues. ELISA: 1:10,000-50,000. Using 50-100ng control peptide/well. Optimal dilutions to be determined by the researcher. Storage and Stability: For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
