- NF kappa B is a heterodimer consisting of a 50kD DNA binding subunit and a 65kD transactivating subunit (RelA). The p50/p65 heterodime is present in the cytoplasm in inactive form bound to its inhibitor, IkB. Upon stimulation of cells with stimuli like LPS, Interleukin-1, IkB gets phosphorylated and degraded by proteosomes releasing the activated NFkB which goes to the nucleus inducing gene expression. NFkB was originally identified as transcriptional activator for kappa light chain gene in B cells. Applications: Suitable for use in Western Blot, Immunofluorescence and Immunohistochemistry. Other applications have not been tested. Recommended Dilutions: Western Blot: 5-10ug/ml for 2 hrs at RT. Immunohistochemistry (Formalin/paraffin): 1:50-1:100 for 20 minutes at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 minutes followed by cooling at RT for 20 minutes. Immunofluorescence (IC): 1:50-1:200 Optimal dilutions to be determined by the researcher. Positive Control: Jurkat cells, Placenta Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
