- Nerve growth factor (NGF) stimulates division and differentiation of sympathetic and embryonic sensory neurons. NGF is thought to have a profound effect on the development and maintenance of sympathetic and embryonic sensory neurons. NGF activity isolated from the male mouse submaxillary gland (MSG) consists of three types of subunits, alpha, beta and gamma, which specifically interact to form a 7S, approximately 130,000-molecular weight (Mr) complex. The 7S complex contains two identical 118-amino acid beta-chains, which are solely responsible for the nerve growth-stimulating activity of NGF. While NGF is found in almost all vertebrates, most research has focused on murine NGF, as the mouse male submaxillary gland contains higher levels of this polypeptide than other tissues. A precursor form of NGF (pro-NGF) selectively binds to p75NTR receptor and leads to apoptosis with expression of TrkA and p75NTR. However, mature NGF binding to TrkA and p75NTR receptors shows opposite effect and leads to activation of cell survival. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 1:100-1:1000 ELISA: 1:100-1:1000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
