- Microfibril-associated glycoprotein (MAGP) is a major structural component of connective tissue microfibrils. MAGP-1 produced by keratinocytes may play some functional role in the formation of dermal matrix organization in the dermis. Applications: Suitable for use in Immunohistochemistry, ELISA, Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: ELISA: 1:1000-1:2000 Western Blot: 1:200-1:500 Immunohistochemistry: 1:50-1:100 Immunoprecipitation: 1:100-1:200 Optimal dilutions to be determined by the researcher. Etching to Enhance Antigenicity: In many cases, antigenicity of elastic fiber components can be enhanced by treating with 6M guanidine HCl, 50mM dithiothreitol, 20mM Tris pH 8.0 for 15 minutes. After washing with 20mM Tris pH 8.0, the slides are treated with 100mM iodoacetamide in the dark for 15 minutes, washed and stained using normal protocols. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS or sterile 50% glycerol, 0.01% Thimerosal. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
