KPNA2 is a 58-60kD member of the Importin alpha family of proteins. It is ubiquitously expressed, and found in both nucleus and cytoplasm. KPNA2 functions as a cargo carrier that transports various complexes from cytoplasm into nucleus. For the MRN DNA repair complex, KPNA2 first binds to NBS1, and then to Importin b1, which mediates movement through the nuclear pore. Human KPNA2 is 529aa in length. It contains an N-terminal IBB/Importin b domain aa2-60, ten Armadillo repeats that bind "cargo" aa71-496 and two intervening NLS binding sites. There is one splice variant that shows a deletion of aa72-310, coupled to an 11aa substitution for aa389-406. Over aa1-132, human KPNA2 shares 94% aa identity with mouse KPNA2. Applications: Suitable for use in Immunocytochemistry. Other applications not tested. Recommended Dilution: Immunocytochemistry: 8-25ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
