- KDM4A (Lysine [K]specific demethylase 4A; also known as JMJD2A) is a155-160kD member of the JHDM3 histone demethylase family of enzymes. It isubiquitously expressed and interacts with the histone deacetylase proteins HDAC1,2, and 3. KDM4A specifically demethylates trimethylated lysines on histone H3 at Lys9 and Lys36. Transcriptional repression occurs when it interacts with NCOR1. Human KDM4A is 1064aa in length (SwissProt #:O75164). It contains one jumonji N domain (aa1355), a catalytic jumonji C domain (aa175291), two PHD zinc finger regions (aa709855), and two TUDOR methylatedhistone binding domains (aa8971011). Over aa386514, human KDM4A shares 93% aa sequence identity with mouse KDM4A. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
