Recommended Dilution: Neutralization of Human IFN-g R1 Bioactivity: The exact concentration of antibody required to neutralize rhIFN-g R1-mediated bioactivity is dependent on the cytokine concentration, cell type, growth conditions and the type of activity studied. The Neutralization Dose50 (ND50) for this antibody is defined as that concentration of antibody required to yield one-half maximal inhibition of the cytokine activity on a responsive cell line, when that cytokine is present at a concentration just high enough to elicit a maximum response. The ND50 is ~0.5-2ug/ml in the presence of 2ng/ml of rhIFN-g, using a confluent culture of HeLa cells. ELISA: 0.5-1ug/ml. Direct ELISA. The detection limit for rhIFN-g R1 is ~0.3ng/well. Western Blot: 0.1-0.2ug/ml. The detection limit for is ~5ng/lane under non-reducing and reducing conditions. Immunohistochemistry: 15ug/ml. Detects IFN-g R1 in human PBMC (peripheral blood mononuclear cells)’s (Fluorescence immunocytochemistry. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
