Insulin Like Growth Factor-I (IGF-I) is a polypeptide growth factor which stimulates the proliferation of a wide range of cell types including muscle, bone, and cartilage tissue. Recombinant Human IGF-I is a single, nonglycosylated, 7.6kD protein containing 70 amino acid residues. Insulin-like growth factor 1 (IGF-I) is involved in regulation of neuronal growth and development in central and peripheral nervous system. It is known to protect neurons against cell death induced by amyloidogenic derivatives, glucose or serum deprivation through pathways involving AKT kinase and transcription factor FKHRL1 phosphorylation. Activation of the insulin-like growth factor system has emerged as a key factor for tumor progression and resistance to apoptosis in many cancers like breast and thyroid cancers. Applications: Suitable for use in Immunohistochemistry, Neutralization, ELISA and Western Blot. Other applications have not been tested. Recommended Dilutions: Neutralization: 0.67-1ug/ml yields one-half maximal inhibition (ND50) of the biological activity of human Insulin-Like Growth Factor I (5ng/ml). ELISA (Sandwich): 0.5-2ug/ml using 100ul/well (antibody solution) In conjunction with Biotinylated anti-human Insulin-Like Growth Factor I (I7661-01B) as a detection antibody, allows the detection of at least 0.2-0.4ng/well of recombinant human Insulin-Like Growth Factor I. Western Blot: 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant human Insulin-Like Growth Factor I is 1.5-3ng/lane, under either reducing or non-reducing conditions. Immunohistochemistry (FFPE): 0.25ug/ml using human breast invasive ductal carcinoma with an overnight incubation at 4ºC. An HRP-labeled polymer detection system was used with a DAB chromogen. Antigen retrival is not required. Optimal dilutions to be detemined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
