- Heparin-binding EGF-like Growth Factor (HB-EGF) is an EGF family growth factor that is synthesized as a transmembrane precursor (proHB-EGF). ProHB-EGF, which contains one EGF-like domain, is subject to variable O-linked glycosylation and N-terminal truncation. In human, this precursor is the cellular receptor for diphtheria toxin. ADAM12 or ADAM17-mediated cleavage of proHB-EGF releases a 22 kD soluble HB-EGF; the cytoplasmic fragment functions as a transcriptional repressor. HB-EGF directly activates EGF R and ErbB4 and promotes the development of many tissues. Human HB-EGF shares 77% aa sequence identity with mouse and rat HB-EGF. Applications: Suitable for use in Western Blot, Neutralization and ELISA. Other applications not tested. Recommended Dilutions: Western Blot: 1ug/ml using recombinant human Heparin Binding EGF-like Growth Factor. Neutralization: ~0.5-2.5ug/ml yields one-half maximal inhibition (ND50) of the biological activity of recombinant human HB-EGF (5ng/ml). Measured by its ability to neutralize HB-EGF-induced proliferation in the Balb/3T3 mouse embryonic fibroblast cell line. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
