- A B Tumor necrosis factor (TNF) and TNF receptor (TNFR) superfamily members are regulators of diverse cellular functions such as proliferation, differentiation, and survival/apoptosis. The glucocorticoid-induced TNFR family-related protein (GITR, also known as TNFRSF18) is an inducible type I transmembrane protein that modulates T cell apoptosis. The human and mouse orthologs share 60% overall aa homology and exhibit three characteristic cysteine rich domains in the extracellular portion of the protein. The intracellular domains of human and mouse GITR are most similar to the TNFR family members 4-1BB, CD27, and OX40. GITR expression is induced on T cells upon activation with various stimuli. The induced expression of GITR appears to confer T cell resistance to anti-CD3 mAb-induced apoptosis. GITR is also expressed on a subset of regulatory T cells (Tregs). In addition, macrophages have also been described as expressing GITR. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ul to label 1x10e6 cells Optimal dilutions to be determined by the researcher. Hybridoma: NSO myeloma cells with B cells from Balb/c mice. Absorption: 620-650nm Emission: 660-670nm Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Caution: APC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
