- Human D-dimer (DD), a specific degradation product of cross-linked fibrin, is released into the bloodstream during clot breakdown. It is used as an indicator of a blood clots, venous thromboses or pulmonary embolisms. Applications: Suitable for use in ELISA and Western Blot. Other applications have not been tested. Recommended Dilution: Sandwich ELISA: Plasma samples should be diluted with 10mM Tris-HCl buffer, pH 7.5, 1M sodium chloride, 0.1% Tween 20. To avoid non-specific binding, the final sodium chloride concentration in plasma samples should be ≥ 0.5M. Western Blot: Non-reducing Optimal dilutions to be determined by the researcher. Hydridoma: Hybridization of Sp2/0 myeloma cells with spleen cells from Balb/c mice. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
