- An intracellular protein termed ERAB (262aa, Endoplasmic reticulum associated Ab binding protein on chromosome X) has been cloned and linked with Ab neurotoxicity. ERAB has structural homology with short-chain alcohol dehydrogenases (hydroxysteroid dehydrogenases and acetyl-CoA reductases). Interestingly, ERAB localizes to endoplasmic reticulum despite the absence of a classical transmembrane sequence. However, in the presence of Ab, ERAB-Ab complex translates to the inner plasma membrane. Alzheimer patients have strong ERAB activity. Normal brains have virtually no ERAB. In addition, Ab neurotoxicity can be reduced by blocking ERAB. Applications: Suitable for use in Western Blot and ELISA. Other applications not tested. Recommended Dilution: Western Blot: 1:1000-1:5000. Detects ~27kD protein in the brain ELISA: 1:10,000-1:100,000 using 50-100ng of control peptide/well. Optimal dilutions to be determined by the researcher. Recommended Control Peptide: E2291-54A: Endoplasmic Reticulum Associated Amyloid Beta Binding Protein, Human, Control Peptide Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
