- Neutrophil Elastase (ELA2), also known as polymorphonuclear leukocyte elastase, is a serine protease belonging to the chymotrypsin family. Located primarily in the azurophil granules of polymorphonuclear leukocytes, ELA2 function is the degradation of many extracellular matrix macromolecules. alpha-1 Antitrypsin (Serpin A1) and secretory leukocyte protease inhibitor (SLPI) have been shown to inhibit ELA2 activity. This protein may be involved in lung emphysema, cystic fibrosis, the adult respiratory distress syndrome (ARDS), rheumatoid arthritis, tumor invasion and infectious diseases. The purified recombinant mouse ELA2 does not contain the last five amino acid residues of the deduced amino acid sequence (NP_056594). Murine myeloma cell line, NSO-derived Ser27-Arg260 with a C-terminal 10-His tag (NP_056594). SDS-PAGE: 34kD, Reducing conditions Specific Activity: ~ 500pmoles/min/ug. Measured by its’ ability to cleave a fluorogenic peptide substrate, MeOSuc-Ala-Ala-Pro-Val-7-amido-4-methylcoumarin (MeOSuc-AAPV-AMC). Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 6 months after receipt at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
