CFTR (Cystic Fibrosis Transmembrane conductance Regulator) is a 170kD chloride (Cl-) channel that regulates salt and water transport across epithelial tissues, where it modulates production of mucus, sweat, saliva, tears, and digestive enzymes. Cystic fibrosis patients have mutations in this protein, resulting in defective in ion transport. Symptoms include reduced transport of sodium chloride (saltier sweat) and digestive and respiratory disorders caused by thicker mucus. CFTR mutations are also associated with congenital bilateral absence of the vas deferens in males. Applications: Suitable for use in ELISA, Western Blot, Immunoprecipitation and Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.5-2ug/ml detects CFTR in RIPA lysates from CFTR-transfected BHK cells. Reported to detect CFTR from a human T84 colon carcinoma epithelial cell lysate. CFTR-transfected BHK cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with C9040-01 (0.5ug/ml). Proteins were visualized using IgG (HRP) goat anti-mouse secondary antibody and a chemiluminescence detection system. Note: Do not boil the lysate. Instead incubate at 37°C for 30 minutes. Immunoprecipitation: Immunoprecipitates CFTR from a T84 cell lysate. Immunocytochemistry: Reported to immunostain CFTR in T84 cells. Note: CFTR runs as a diffuse protein on SDS-PAGE. Optimal dilutions to be determined by the researcher. Recommended secondary antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-06H: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
