- CD11b (integrin alphaM subunit) combines with CD18 (integrin beta2 subunit) to form the integrin Mac-1, also known as complement receptor 3 (CR3), which mediates adhesion to C3bi and ICAM-1 (CD54).2-6 Mac-1 is expressed at varying levels on granulocytes, macrophages, dendritic cells, NK cells, and B-1 cells in the peritoneal and pleural cavities.2, 7-10 In addition to its role in binding C3bi on opsonized targets and mediation of the subsequent ingestion process,4, 11 Mac-1 is important as an adhesion molecule in the transendothelial migration of monocytes and neutrophils.12 Applications: • Flow cytometry 1 • Immunohistochemistry (frozen sections) 13 • Immunoprecipitation 1 Recommended Dilution: Flow Cytometry: ≤ 0.3ug/10e6 cells Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Store at -20°C or colder. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
