- Porcine CD8, a type I transmembrane glycoprotein, is expressed as either a CD8alpha/alpha homodimer or a CD8alpha/beta heterodimer. It is found on most thymocytes, and on the “suppressor/cytotoxic” subpopulation of peripheral T cells. CD8 functions primarily as a co-receptor with MHC Class I-restricted T-cell receptors in antigen recognition. Applications: • Flow cytometry1-3 • Immunoprecipitation1 • Cytotoxicity assays1,2 Recommended Dilution: Flow Cytometry: ≤ 0.2ug/10e6 cells Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Store at -20°C or colder. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. PE/CY5 combination conjugate is used as a third fluorochrome in multicolor flow cytometry applications when only a single 488nm excitation source is available. Antibodies are covalently coupled using heterobifunctional linking reagents, resulting in an optimal conjugation format for flow cytometry.
