- Water is a critical component of all living cells. Interestingly, tissue membranes show a great degree of water permeability. Mammalian red cells, renal proximal tubules, and descending thin limb of Henle are extraordinarily permeable to water. Water crosses hydrophobic plasma membranes either by simple diffusion or through a facilitative transport mechanism mediated by special protein “aquaporin”. Over the last decade, genes for several members of aquaporin family (AQP1-9 and AQPAP) have been cloned, expressed, and their distribution studied in many tissues. AQP2 (AQP-CD or WCH-CD; 271aa; human gene locus 12q13) is the vasopressin-regulated water channel of the apical membrane of collecting duct cells. AQP families of proteins are predicted to contain six transmembrane domains. The N- and C-termini are predicted to be cytoplasmic. Defects in AQP2 are a cause of an autosomal dominant form of nephrogenic diabetes insipidus (NDI). Applications: Suitable for use in ELISA. Western Blot, though not tested, may potentially be used as an application. Other applications not tested. Recommended Dilutions: ELISA: 1:10,000-1:50,000; Coat plates with control peptide at 1ug/ml. Optimal dilutions to be determined by the researcher. Recommended Control Peptide: A3000-07M: Aquaporin 2, Rat Control Peptide Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
