- The purine nucleoside adenosine modulates diverse physiological functions including induction of sedation, vasodilatation, suppression of cardiac rate and contractility, neurotransmitter release, inhibition of platelet aggregation and lipolysis. Adenosine released from cells interacts with membrane receptors (adenosine receptors, ARs). Based upon biochemical and pharmacological criteria, ARs have been classified into A1, A2a, A2b and A3. The high affinity receptor A1 inhibits adenyl cyclase via G proteins. A2a receptor is believed to cause vasorelaxation in the coronary artery. Various ARs and their subtypes have recently been cloned from several species. ARs belong to the superfamily of G-protein coupled receptors and predicted to contain 7 transmembrane domains. The N-termini and C-termini are predicted to be extracellular and cytoplasmic, respectively. There is an overall 30% homology among the four ARs. ARs are distributed throughout the body. Gene location of various ARs: A1 (Chromosome 1; q.31.3-32.2); A2a (Chromosome 22); A2b (Chromosome 17; p.11.2-12); A3 (Chromosome 1). Applications: Suitable for use in ELISA. Western Blot, though not tested, may potentially be used as an application. Other applications not tested. Recommended Dilution: Western Blot: 1:1000-1:5000 ELISA: 1:10,000-1:50,000 using 50-100ng A3R control peptide per well. Optimal dilutions to be determined by the researcher. Control Peptide: A0869-70G Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

- Recognizes Adenosine A3 Receptor. Species crossreactivity: The peptide used for making antibodies to rat A3R shows 46% homology (7/15 AA) with human and 20% (3/15 AA) with sheep A3R sequences. Antibody crossreactivity in various species is not established.
