- The current monkeypox strain has approximately 50 genetic variations compared to related viruses that circulated in 2018-2019. While PCR methods are very accurate, they are vulnerable to genetic mutations as they only detect a short sequence within the genome. In contrast, serologic assays are less sensitive to mutational changes due to the high degree of antigenic relation among related virus strains and the polyclonal nature of the immune response. Serologic assays provided significant diagnostic support during the 2003 U.S. monkeypox outbreak. Monkeypox virus-specific IgM is detectable within 2 days of rash onset and monkeypox IgM capture assays have been shown to distinguish between vaccinated and unvaccinated individuals with a sensitivity and specificity of 95% (doi:10.1128/CDLI.12.7.867-872.2005). Represents immunodominant regions of the major virion core protein. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70°C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
