- Palladin is a component of actin-containing microfilaments that control cell shape, adhesion, and contraction. The deduced 789aa mouse protein contains an N-terminal serine- and proline-rich region and 3 C-terminal IgC2 domains. Palladin was expressed as a 90- to 92kD doublet in fibroblasts. Northern and Western blot analysis of chick and mouse tissues detected multiple isoforms. Expression was ubiquitous in embryonic tissues and was downregulated in certain adult mouse tissues. Palladin colocalized with alpha-actinin in stress fibers, focal adhesions, cell-cell junctions, and in embryonic Z lines. The 2 proteins coimmunoprecipitated from fibroblast lysates. The deduced 772aa protein shares about 31% identity over 448aa with chicken connectin-1. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.01-2ug/ml Immunohistochemistry: 5-20ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
