- Retinoic acid mediates tissue-specific expression of target genes through its binding to nuclear hormone receptors. To activate transcription, these receptors bind specific sites, called hormone response elements, within the target gene's regulatory region. Willy et al. identified a cDNA encoding NR1H3, or LXRA, an orphan member of the nuclear receptor superfamily. The predicted 447aa LXRA protein contains a DNA-binding domain and a putative ligand-binding domain; the aa sequences of these domains are 77% identical to those of NR1H2. Willy et al. identified a distinct retinoid response element for LXRA/RXRA heterodimers, termed the LXR-responsive element, that consists of 2 degenerate copies of the consensus hexad sequence spaced by 4 nucleotides. Applications: Suitable for use in ELISA, Western Blot, Immunohistochemistry, Immunocytochemistry. Other applications not tested. Recommended Dilution: ELISA: 1:100-5000 Western Blot: 1:50-400 Immunohistochemistry (frozen): 1:50-500 Immunohistochemistry (paraffin): 1:10-100 Immunocytochemistry: 1:50-500 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
