Glutamate-cysteine ligase, also known as gamma-glutamylcysteine synthetase, is the first rate limiting enzyme of glutathione synthesis. The enzyme consists of two subunits, a heavy catalytic subunit and a light regulatory subunit. Gamma glutamylcysteine synthetase deficiency has been implicated in some forms of hemolytic anemia. It catalyzes the ATP-dependent condensation of cysteine and glutamate to form the dipeptide gamma-glutamylcysteine. The peptide bond in this peptide product is between the carboxylate group of the glutamate and the amino group of the cysteine. The product of the GCL-mediated condensation reaction of L-glutamate and L-cysteine is gamma-glutamylcysteine, which is readily condensed with glycine, by glutathione synthetase, to form glutathione. Applications: Suitable for use in Western Blot, Immunohistochemistry, Immunocytochemistry and Immunofluorescence. Other applications not tested. Recommended Dilution: Western Blot: 0.5-2ug/ml Immunohistochemistry: 5-20ug/ml Immunofluorescence (IC): 5-20ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
