- MaxLight™650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000. Recognizes a conserved determinant of the p53 protein. Its epitope maps near the C-terminus end (aa 370-378) of p53. It has been used successfully to immunoprecipitate p53 oncoprotein complexes with simian virus 40 (SV40) large T antigen. Reportedly, this antibody precipitated the most ATPase-active form of T. p53 is a tumor suppressor gene expressed in a wide variety of tissue types and is involved in regulating cell growth, replication, and apoptosis. It binds to MDM2, SV40 T antigen and human papilloma virus E6 protein. Mutations involving p53 are found in a wide variety of malignant tumors, including breast, ovarian, bladder, colon, lung, and melanoma. Applications: Suitable for use in Immunofluorescence, FLISA, Flow Cytometry, Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 0.5-1ug/million cells Immunofluorescence: 0.5-1ug/ml Western Blot: 0.5-1.0ug/ml Immunoprecipitation: 0.5-1ug/500ug protein lysate Immunohistochemistry: Frozen only: 0.5-1.0ug/ml for 30 minutes at RT Optimal dilutions to be determined by the researcher. Positive Control: MDA-MB-231 Cells. Breast or Colon carcinoma Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
