- GALNT2 (NAcetylGalactoseaminyl Transferase 2; also UDPAcetylgalactosaminyltransferase 2 and ppGalNAcT2) is a 6874kD member of the GalNAC transferase subfamily, glycosyltransferase 2 family of enzymes. It is widely expressed, being found on basal keratinocytes, hepatocytes, B cells, renal tubular epithelium, and virtually all cell lines examined to date. GALNT2 is found in the Golgi apparatus, and catalyzes the transfer of UDPGalNAC onto either a Ser or Thr residue on a previously glycosylated peptide/polypeptide backbone. The generation of Olinked carbohydrates is believed to play a role in cytokine proteolytic processing, as the presence of Olinked sugar adjacent to a PC processing site is known to inhibit proteolysis and molecule inactivation. Human GALNT2 is a 571 amino acid (aa) type II transmembrane protein. It contains a six aa Nterminal cytoplasmic region and a 547aa extracellular domain (aa25571). The ECD possesses two key parts, a catalytic region with two catalytic subdomains (aa135240 and 300362), and a ricintype lectin domain that binds carbohydrates (aa456566). The latter domain is suggested to facilitate GALNT2 action by imparting specificity and stability to the overall enzyme activity. A 52kD soluble form of GALNT2 has been reported that begins at Lys52. There are two potential splice form variants. Both contain a four aa substitution for aa142, and one contains an additional four aa substitution for aa543571. Over aa52571, human GALNT2 shares 97% aa sequence identity with mouse GALNT2. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O or PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
