- Contactin-2 (CNTN2), also called TAG-1 (transient axonal glycoprotein), TAX1 (transiently-expressed axonal glycoprotein), or axonin-1, is a 135kD glycosyl‑phosphatidylinositol (GPI)- anchored cell adhesion molecule that belongs to the contactin subfamily within the immunoglobulin (Ig) protein superfamily. Human Contactin-2 cDNA encodes a 28 amino acid (aa) signal peptide, a 984aa mature secreted protein with six Ig-like domains followed by four fibronectin type III‑like repeats, and a 28 aa C-terminal GPI anchor pro-sequence. GPI-specific phospholipase activity can release soluble, active Contactin-2 from the membrane. Mature human Contactin-2 shares approximately 93%, 93% and 75% aa sequence identity with human, rat and chicken Contactin-2, respectively. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1ug/ml with human cerebellum tissue under reducing conditions. Immunohistochemistry: 5-15ug/ml in immersion fixed paraffin-embedded sections of human brain (cortex). Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
