- Retinal-specific ATP-binding cassette transporter encoded by the gene ABCA4 plays a critical role in the transport of retinoid substrates from the extracellular or intradiscal rod cell membranes to the cytoplasmic membrane surface for processing ultimately into vitamin A and then transferred to the retinal pigment epithelium where it is converted to 11-cis-retinal that active component needed for vision. ABCA4 is localized to the outer segments of rods and cones with a million copies per photoreceptor. ABCA4 is retinal specific protein. Mutations in ABCA4 are linked to several eye-related genetic diseases, such as forms of macular degeneration called Stargardt disease and ARMD2, as well as Retinitis pigmentosa 19, and Cone-rod dystrophy 3. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Immunohistochemistry (paraffin): 1:250 detected ABCA4 in human retina tissue. Requires HIER. Western Blot: 5ug/ml detected ABCA4 in 200ug of Y79 cell lysate. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
