- S1P5 is also known as EDG-8 and nerve growth factor-related G-protein-coupled receptor-1 (NRG-1). S1P5 is a 398 amino acid (aa) seven-transmembrane receptor putative glycoprotein that binds the lysolipid phosphoric acid mediator, sphingosine 1-phosphate. Extracellular portions of human S1P5 show 96% and 97% aa identity with mouse and rat S1P5, respectively. Isoform 1 is expressed at a low level in peripheral tissues. Isoform 2 has an alternate C-terminal that is 88 aa shorter and is expressed mainly in brain, spleen, and PBMC. S1P5 is upregulated in large granular lymphocytic leukemias. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 0.25ul labels 1x10e6 cells. Validated using HEK293 human embryonic kidney cell line transfected with human EDG8 and eGFP. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
