Alpha-synuclein (A Alpha-synuclein), a neuronal protein encoded by the SNCA gene, plays a central role in synaptic function and is a key pathological driver in Parkinson's disease and related synucleinopathies. Aggregation of A Alpha-synuclein into fibrillar structures is a hallmark of disease progression, contributing to the formation of Lewy bodies and neuronal dysfunction. Biotinylated A Alpha-synuclein pre-formed fibrils (PFFs) are chemically tagged with biotin, enabling precise detection, immobilization, and tracking in experimental systems. These modified fibrils retain the structural and functional characteristics of native A Alpha-synuclein aggregates, making them highly valuable for studying the mechanisms of fibril propagation, cellular uptake, and seeding of endogenous A Alpha-synuclein. In neurodegenerative disease models, biotinylated PFFs facilitate high-resolution analysis of intercellular transmission, aggregate-induced toxicity, and protein clearance pathways. Their compatibility with streptavidin-based imaging and capture techniques allows for enhanced visualization and quantification of pathological processes in vitro and in vivo. Biotinylated A Alpha-synuclein PFFs serve as a robust platform for screening therapeutic candidates aimed at inhibiting fibril formation, blocking propagation, and restoring neuronal health. Their use accelerates translational research by providing reproducible and disease-relevant tools for understanding the molecular basis of synucleinopathies. A 15 amino acid tag on the C-terminal tail of Alpha Synuclein facilitates site-specific covalent biotinylation (1).
