- Tau protein, encoded by the MAPT gene (UniProt ID: P10636), is essential for stabilizing microtubules and maintaining neuronal architecture. The Tau-441 isoform (2N4R), the longest variant expressed in the adult human brain, contains two N-terminal inserts and four microtubule-binding repeats, making it a key model for studying tauopathies. While monomeric Tau performs vital cellular functions, under pathological conditions it can misfold and assemble into soluble oligomers-intermediates in the aggregation pathway leading to neurofibrillary tangles. Wild-type Tau-441 oligomers are increasingly recognized as neurotoxic species that disrupt synaptic function, impair axonal transport, and trigger inflammatory responses, even in the absence of mutations. Tau-441 wild-type oligomers are used in experimental systems to investigate early-stage tauopathy mechanisms, including conformational changes, post-translational modifications, and prion-like propagation.
