- Amyloid beta (AB Beta) peptides, particularly the pyroglutamate-modified variant AB Beta 3-42, are increasingly recognized as key contributors to Alzheimer's disease. This truncated form arises from post-translational modification of AB Beta peptides, where the N-terminal glutamate is cyclized into pyroglutamate, enhancing hydrophobicity, resistance to degradation, and aggregation propensity. Pre-formed fibrils (PFFs) of AB Beta 3-42 replicate the B Beta-sheet-rich architecture of amyloid plaques found in Alzheimer's brains. These fibrils exhibit heightened neurotoxicity compared to full-length AB Beta 1-42, due to their increased stability and ability to seed further aggregation. AB Beta 3-42 PFFs are particularly effective at inducing synaptic dysfunction, neuroinflammation, and neuronal death, making them a powerful model for studying the molecular mechanisms of disease progression.
