Tau protein, encoded by the MAPT gene (UniProt ID: P10636), is a microtubule-associated protein essential for maintaining neuronal structure and intracellular transport. The Tau-430 isoform (2N4R) includes two N-terminal inserts and four microtubule-binding repeats, representing a key variant expressed in the adult human brain. The P290S mutation, located within the microtubule-binding domain, alters Tau's conformation and reduces its affinity for microtubules. This destabilization promotes early misfolding events, making the monomeric form of Tau-430 P290S a valuable model for studying the initial stages of tauopathy. Unlike aggregated fibrils, mutant monomers provide insight into the pre-aggregation phase, where subtle biochemical changes begin to disrupt neuronal homeostasis.
