Tau protein, encoded by the MAPT gene (UniProt ID: P10636), is a microtubule-associated protein essential for maintaining neuronal structure and function. The Tau-441 isoform (2N4R) is the longest adult brain variant, containing two N-terminal inserts and four microtubule-binding repeats, making it highly relevant to tauopathy studies. The P301S mutation, located within the microtubule-binding domain, enhances Tau's tendency to misfold and aggregate into B Beta-sheet-rich fibrils. Pre-formed fibrils (PFFs) generated from Tau-441 P301S mutant protein replicate key pathological features of neurodegenerative diseases such as Alzheimer's disease and frontotemporal dementia, including the formation of neurofibrillary tangles and prion-like propagation of Tau aggregates. These mutant PFFs are widely used in cellular and animal models to investigate the mechanisms of Tau seeding, neurotoxicity, and intercellular transmission. Their structural fidelity to human pathological Tau makes them ideal for evaluating therapeutic strategies aimed at inhibiting aggregation, enhancing clearance, or blocking spread. Tau-441 P301S PFFs are instrumental in advancing biomarker discovery and drug development, offering a reproducible and disease-relevant platform for translational neuroscience.
