- Tau protein, encoded by the MAPT gene (UniProt ID: P10636), is a microtubule-associated protein essential for maintaining neuronal structure and function. The Tau-441 isoform (2N4R) is the longest adult brain variant, containing two N-terminal inserts and four microtubule-binding repeats, making it highly relevant to tauopathy studies. The P301S mutation, located within the microtubule-binding domain, enhances Tau's tendency to misfold and aggregate into B Beta-sheet-rich fibrils. Pre-formed fibrils (PFFs) generated from Tau-441 P301S mutant protein replicate key pathological features of neurodegenerative diseases such as Alzheimer's disease and frontotemporal dementia, including the formation of neurofibrillary tangles and prion-like propagation of Tau aggregates.
