The GABA-A receptor alpha 5 subunit (GABRA5) is a critical component of the GABA-A receptor complex, the brain's primary fast-acting inhibitory neurotransmitter system. As part of the ligand-gated ion channel superfamily, GABRA5 is predominantly expressed in the hippocampus, where it plays a central role in modulating tonic inhibition, synaptic plasticity, and cognitive function. Recent research has highlighted GABRA5 as a promising target in neurodegenerative disease, particularly Alzheimer's disease. Inverse agonists selective for GABRA5-containing receptors have shown potential in enhancing learning and memory by reducing excessive inhibitory tone, which is often elevated in neurodegenerative states.
