The Cav Beta 2 subunit is a critical auxiliary component of voltage-gated calcium channels (VGCCs), playing a central role in neuronal excitability and synaptic signaling. It facilitates the trafficking of the pore-forming alpha1 subunit to the plasma membrane by masking an endoplasmic reticulum (ER) retention signal, ensuring proper channel localization and surface expression. This process is essential for the functional assembly of VGCCs at the neuronal membrane. Beyond trafficking, Cav Beta 2 directly influences the biophysical properties of calcium channels, including voltage sensitivity, activation kinetics, and inactivation dynamics.
