Cav1.3, encoded by the CACNA1D gene, is a voltage-gated L-type calcium channel subunit predominantly expressed in neurons and neuroendocrine cells. It plays a vital role in regulating calcium influx, which is essential for neuronal excitability, synaptic plasticity, gene expression, and neurosecretion. In the central nervous system, Cav1.3 contributes to pacemaker activity in dopaminergic neurons and modulates long-term potentiation-key processes underlying learning and memory. Dysregulation of Cav1.3-mediated calcium signaling has been implicated in several neurodegenerative and neuropsychiatric disorders, including Parkinson's disease, Alzheimer's disease, and autism spectrum disorders. Its role in dopaminergic neuron vulnerability makes it a potential therapeutic target in Parkinsonian syndromes. Beyond the brain, Cav1.3 is also expressed in the cochlea, where it is essential for auditory transduction in inner hair cells and electromotility in outer hair cells-highlighting its importance in sensory processing. Additionally, its presence in cardiac atrial tissue suggests a role in atrial excitability and arrhythmogenesis, particularly in atrial fibrillation. Given its broad physiological roles and emerging links to neurological disease, Cav1.3 is a critical focus in neuroscience and neurodegeneration research. Targeting Cav1.3 may offer novel strategies for modulating calcium-dependent signaling pathways in both central and peripheral disorders.
