- Wortmannin is a fungal metabolite that irreversibly inhibits phosphatidylinositol 3-kinase (PI3K), a key regulator of intracellular signaling pathways involved in growth, metabolism, and autophagy. In neuroscience, Wortmannin is used to investigate the PI3K/Akt/mTOR pathway, which plays a central role in neuronal survival, synaptic plasticity, and neuroinflammation. By inhibiting PI3K, Wortmannin suppresses autophagy and disrupts membrane trafficking-processes essential for neuronal maintenance and function. Its application in neurodegenerative disease models helps elucidate the impact of impaired PI3K signaling on protein aggregation, mitochondrial dysfunction, and cell death.
